1. Pharmacokinetics
Esomeprazole:
Absorption: Rapid absorption, peak plasma concentration within 1–2 hours.
Metabolism: Metabolized in the liver by the P450 system.
Half-life: Approximately 1–1.5 hours.
Domperidone:
Absorption: Well absorbed, with reduced bioavailability due to first-pass metabolism.
Half-life: Approximately 7–9 hours.
2. Pharmacodynamics
Esomeprazole: Proton pump inhibitor, reduces stomach acid production.
Domperidone: Dopamine antagonist, enhances gastric motility.
3. Clinical Indications
GERD: Treatment of heartburn and esophagitis.
Peptic Ulcers: Healing and prevention.
Delayed Gastric Emptying: Reduces symptoms of nausea, vomiting, and bloating.
Nausea & Vomiting: Effective in controlling nausea and vomiting.
4. Dosage and Administration
Dosage: 1 capsule (40mg Esomeprazole & 30mg Domperidone) once daily before meals.
Administration: Take 30 minutes before meals, swallowing the capsule whole.
5. Precautions
Allergy: Contraindicated in patients with hypersensitivity to Esomeprazole, Domperidone, or any components of the formulation.
Cardiac Risk: Use cautiously in patients with heart conditions due to potential QT interval prolongation.
